It optically combines beams of radiation from tungsten and deuterium lamps and sends them through a sample. The beam exits the sample and is sent via a grating to a photodiode array which analyzes multiple wavelengths simultaneously. The absorbance spectrum generated can be used to identify the components of the sample and their concentrations in the sample mixture. Directions for Use General Instructions for Use. Turn the UV-VIS on and allow it to warm up. Start ChemStation software in 'online' mode. Under the Task pulldown menu, select Fixed Wavelengths and enter the wavelength of interest in the Use wavelengths field.

Place the cell containing the water in the cell compartment and tighten the clamp. Click on Blank. Release the clamp and remove the cell. Rinse and fill the cell with your sample solution. Place the cell in the compartment. Click on Sample in the ChemStation window. When the window showing the spectrum appears, type in an identifying name.

Tips for Use. Warning: There is no one person responsible for the upkeep of these instruments. As such, it is your responsibility to insure that the UV-VIS you are using is working properly and is set up correctly.

Note: The 8452 in Friedland 202 needs to be reblanked after every 15-20 minutes of use!. Note: Allow a UV-VIS to warm up for about 30 minutes before first use for best results. If you will be using it repeatedly over a few hours, just leave it on, rather than starting up and shutting down repeatedly. The cuvette holder breaks somewhat easily. Be sure to open the holder using the lever.

When loading the cuvette, insure that the clear sides are open to the lamp and that there are no air bubbles in the path, which will skew the reading. If you find that the sample peaks are moving/changing for multiple runs using the same sample, you need to reblank the instrument. Click on the 'Blank' button in the ChemStation software to reblank. The UV-VIS will click 3 times and then be ready for use again.

Follow proper procedure for starting up. The indicator on the instrument will read 'Busy' while it is warming up. Once 'Power' and 'Lamp' are displayed in green, the instrument has warmed up and is ready for use. (Also see Note above.) Also, be sure to start the ChemStation software on the PC in 'online' mode. Solutions to be analyzed can be any color but they must be clear. If you are getting odd spectra, insure that the Deuterium lamp is on. Check the Instrument Panel in the ChemStation software to check lamp configuration.
